Product Description
The kit # 27186 has improved stability and convenience from the former kit # 17186. The improved kit has some advantages as follows. 1) We have changed the formulation of HRP conjugated antibody from lyophilized product to liquid (0.4mL X 1). By this change, the stability of the conjugate has improved so that customers can be use it for 12 months even in stored at 2-8C and can use it several times. 2) The assay range has changed from 2.15 - 275 ng/mL to 1.09 - 70 ng/mL (sensitivity is 0.25 ng/mL). Additionally, we recommend the customers to use IBLysis-I (# 19022) instead of TNE buffer for sample treatment. IBLysis-I was developed by improving TNE buffer solution for solubilization of cultured cells and tissue. It is ready to use for pretreatment of samples. Cyclooxygenase (COX) is a membrane bound enzyme responsible for the oxidation of arachidonic acid to Prostaglandin G2 (PGG2) and the subsequent reduction of PGG2 to PHG2. These reactions are the first steps in the formation of a variety of prostanoids. COX has been shown to be expressed in at least two different isoforms, a constitutively expressed form, COX-1, and an inducible form, COX-2. COX-1 is thought to regulate a number of housekeeping functions, such as vascular hemostasis, renal blood flow, and maintenance of glomerular function. Inflammation mediators such as growth factors, cytokines and endotoxin induce COX-2 expression in a number of cellular systems.

Q.Can I re-use standard after reconstitution?

A.Not recommended to re-use standard after reconstitution. Please use it at once after the reconstitution.
Please note that there are some exceptions. One time freeze-thaw the standard is acceptable for use after reconstitution for some ELISAs.

Q.What is different between reagent blank and test sample blank?

A.Reagent blank means a well is only added EIA buffer and the purpose is confirming whether the Test sample value is influenced by lack of washing process or other operations. Test sample blank means a well is added EIA buffer and HRP antibody and the purpose is to calculate the background.

A.The pre-coated plate contained in our ELISA kit is 96 wells plate. We recommend to use 16 wells (2 slits) for standard and 80 wells (10 slits) for 40 samples in duplicate.

Q.How to wash an ELISA plate?

A.Washing it by an auto-washer is highly recommended.

Q.The wells turned black during the test with the kit.

A.It is possible that the wells were not washed sufficiently during the washing process after the HRP-labeled antibody reaction.
Be sure to wash the wells enough times as described in the data sheet with washing buffer of more than 350 µL.
