Q.Does the method have any correlation with existing standard method?

A.The method is correlated with ultracentrifugation method.

Q.How samples should be stored until the shipping?
-
A.Please keep samples at -80C. Although one freeze-thaw cycle is acceptable, the repeated cycles will affect the data.

Q.Does hemolysis affect the results?
-
A.A clear peak appears for hemolysis after HDL and the peak usually does not affect the calculation of HDL data.

Q.Is there any requirement for feeding animals or subjects?
-
A.The method has no particular requirement for the way of feeding. It depends on the study.

Q.Is there any validation data such as CV data and reproducibility?
-
A.Various validation data are available.

Q.Can you provide the fraction of lipoprotein?
-
A.Unfortunately, it cannot be provided. LipoSEARCH is a HPLC based lipoprotein analyzing service for providing the concentration, size of 4 major classes and its sub-classes in details through the process of lipoprotein separation and enzymatic reaction by a post column. We do not provide any services for separating and collecting lipoprotein.

Q.What kind of plasma sample can be used?
-
A.Any plasma samples (EDTA-plasma, citrate-plasma and heparin-plasma) which are collected by blood collection tubes coated anticoagulants can be used.
However, postheparin plasma (intravenous heparin) cannot be used. -
Q.Is cell culture supernatant applicable for analysis by LipoSEARCH?
-
A.Yes. The volume is required more than 250µL for the analysis.

Q.Is it possible to analyze cell culture supernatants in 96-well plates?
-
A.No it is not applicable. Please prepare your samples in micro tubes.

Q.Can samples derived from genetically modified cells be analyzed by LipoSEARCH?
-
A.No. Such samples (including virus-infected cells) are not acceptable.

Q.Can samples inactivated by surfactants be analyzed by LipoSEARCH?
-
A.No. because it is difficult to obtain accurate data with the samples containing surfactants.
There is also a risk for damaging the analytical system if we run the system with such samples.

Q.How many samples can be measured?
-
A.It depends on type of automated analyzers.
For example, 50 samples can be measured using by Tokyo Boeki Biolis24iP type automated analyzer. -
Q.Can this kit measure LPL/HTGL activity using LPL-deficient patient sample?
-
A.As it is considered that the level of LPL activity is extremely low in LPL-deficient patient sample, it can be lower than LLOQ (Lower Limit of Quantification)and also possibly not detect.
We highly recommend for validation of sensitivity of your auto-analyzer prior sample measurement. -
Q.Is there any information regarding the stability of this kit in EDTA-plasma sample?

A.Yes, there is. We confirmed that there was no significant difference between the value of fresh samples (soon after the blood collection) and the samples stored at 4 °C for 2 days.
It was also no significant issue after freeze thaw. -
Q.Has the stability of the reagents of this kit on-board been evaluated?
-
A.Yes it has. In the result, the CV was less than 20% in comparison between the reagents on-board (refrigerated compartment) of a clinical chemistry analyzer and the opened reagents stored at 2-8℃ in 28 days.

Q.Can this kit be used for animal samples?
-
A.Yes, it can. It can be used for mouse, rat and rabbit plasma samples after administration of heparin and LPL and HTGL activity can be detected as same as human samples.

Q.Is it possible to use Neosilk®-Human Type Collagen I as a cell culture scaffold?
-
A.It can be used as the coating material for cell culture dishes. Because it does not contain any animal-derived ingredients, it can be used for xeno-free cultures. However, Neosilk®-Human Type Collagen I does not have triple-helical structure, and for this reason there are times the adhesive ratio is low for certain cell types. In addition, it does not form a collagen gel, so it cannot be used for collagen gel cultures.

Q.Are proteins produced from transgenic silkworms subject to the Cartagena protocol?

A.No they are not. Because the transgenic silkworm-based protein production system does not use viruses, the host organisms subject to the Cartagena protocol are the silkworms. The proteins extracted from the cocoons do not contain the host organism (transgenic silkworm), so they are not subject to the Cartagena protocol.

Q.Does Neosilk®-Human Type Collagen I have a triple-helical structure?

A.No, it consists of a single α1(I) chain.
